In Madagascar: Operation Penguin, fans play as the crafty penguins, the breakout stars of the movie Madagascar. Taking on the role of Private, gamers embark on a single-player, 2D side-scrolling, action-packed journey leading a daredevil zoo escape to Antarctica and the wild island of Madagascar. Along the way, players utilize inventive secret agent gadgets and conquer a host of challenging tasks in 18 mission-based chapters.

- Lots of Wild Gadgets – Gamers use crazy contraptions and unique gadgets, such as a soda bottle jet pack or hamburger wrapper parachute, to help break free from the confines of the zoo. They can climb "flipper over flipper" using candy canes as hooks, hurl fruit with a homemade slingshot and launch sleeper darts through a twisty straw. They must also defeat enemies with a secret "bass knuckles" weapon – a sea bass that serves as a dastardly device.
- Meet Up With Movie Star Mammals – Film favorites Alex the Lion, Marty the Zebra, Melman the Giraffe and Gloria the Hippo each get their own special level in Madagascar: Operation Penguin. Playing as Alex, fans roar and jump to battle monkeys. On Marty level, they pull off a tough stealth mission. Melman takes the lead in a dancing mini-game, and Gloria gets her own level where a case of stolen fruit gets her hopping mad.
- Escape To Exotic Lands – Players adventure through 18 mission-based chapters while battling enemies, collecting useful tools, scouting out danger and exploring new surroundings. They can also travel across zoo environments like the Lion Pen and Monkey Cage, and take a dangerous trek through the subway system, hop on a boat bound for Africa and explore Antarctica and Madagascar. Additionally, players assist zoo mates along the way to earn their help in later missions and they can navigate freely between completed levels for even more replayability.
- Bust New Moves Against A Boatload Of Enemies – Players run, jump, karate chop and swim along the water surface while trying to escape enemies such as foosa, pigeons, rats, bees, porcupines, chameleons, baboons, snakes, walruses, crocodiles and lemmings. They can also power-up with special moves and employ the crafty "Smile and Wave" to outwit foes.
